Resveratrol with antioxidant activity inhibits matrix metalloproteinase via modulation of SIRT1 in human fibrosarcoma cells.

Abstract:

AIMS: Resveratrol, a silent information regulator 1 (SIRT1) activator, has been reported to act as an antioxidant contained in red wine and prevent the development of cardiovascular diseases. Histone deacetylase such as SIRT1 is involved in the regulation of lifespan extension. In this study, the effect of resveratrol on matrix metalloproteinases that play an important role in metastasis was examined in human fibrosarcoma cell line, HT1080.
MAIN METHODS: The effect of resveratrol on MMPs activity was evaluated using gelatin zymography. Western blot analysis and RT-PCR assay were used to determine the effect of resveratrol on the expression level of MMP-9, MAPK and SIRT1 proteins and genes, respectively.
KEY FINDINGS: It was observed that resveratrol exhibited not only antioxidant effects on lipid peroxidation and DNA oxidation but also inhibitory effects on the expression of MMP-2 and 9 in HT1080 cells stimulated with either phorbol myristate acetate or phenazine methosulfate. Furthermore, it was found that treatment with resveratrol decreased the level of MMP-9 expression via down-regulation of p-ERK, c-fos and p65. In addition, the level of SIRT1 gene expression was also enhanced by treatment of resveratrol alone but the level of MMP-9 gene expression was decreased.
SIGNIFICANCE: These results suggest that the activation of SIRT1 in the presence of resveratrol especially inhibits the expression of MMP-9 in HT1080 cells, providing an evidence that resveratrol can be a potential candidate for chemoprevention of cancer.

El resveratrol, un regulador de (SIRT1), se ha visto que actua como un antioxidante contenido en el vino tinto y previene el desarrollo de enfermedades cardiovasculares. La SIRT1 está implicada en la regulación de la extensión de la vida útil. En este estudio, el efecto del resveratrol en metaloproteinasas de la matriz que desempeñan un importante papel en la metástasis se examinó en la línea de células humanas de fibrosarcoma, HT1080.
El efecto del resveratrol sobre la actividad de las MMPs se evaluó Western blot y RT-PCR que se utilizaron para determinar el efecto del resveratrol en el nivel de expresión de MMP-9, MAPK y SIRT1sus proteínas y genes, respectivamente.
Se observó que el resveratrol mostró no sólo los efectos antioxidantes sobre la peroxidación de lípidos y la oxidación del ADN, sino también efectos inhibitorios sobre la expresión de MMP-2 y 9 en las células HT1080 estimulados con acetato de forbol miristato bien o metosulfato fenazina. Además, se encontró que el tratamiento con resveratrol redujo el nivel de expresión de MMP-9 a través de la baja regulación de p-ERK, c-fos y p65. Además, el nivel de expresión del gen SIRT1 también fue mejorado por el tratamiento de resveratrol solo, pero el nivel de expresión del gen MMP-9.
Estos resultados sugieren que la activación de SIRT1 en presencia de resveratrol en particular inhibe la expresión de MMP-9 en las células HT1080, proporcionando una evidencia de que el resveratrol puede ser un candidato potencial para la quimioprevención del cáncer.
